I have read that the DOSBox Daum compile has share support, but I think the directory/file system caching will still give issues.
The share support I've seen is within the same process only. Not saying they haven't extended it, but I haven't seen that.
Windows builds of DOSBox appear to support share modes now... by using the share mode support in Windows. Linux builds of DOSBox do not.
DOSEmu appears to have almost complete share support for redirected Linux volumes (not disk images). The differences are subtle, but could still cause problems with many multinode doors due to fcntl() flock insanity. If you want to audit a door for compatability, the big thing to watch out for is for a locked file to be open more than once at the same time. If this ever occurs, the door cannot safely be ran multinode with dosemu. If it doesn't occur, you should be safe... the other problems are likely to prevent the door from working at all.
Easiest way to check would be to trace syscalls... strace(1) would be the tool to test doors with.
